Clinical Reference Monograph: Oxycodone & Acetaminophen 10mg/325mg (Percocet E712)
Compiled from official US FDA Prescribing Information, DailyMed Labeling & Clinical Guide • DEA Schedule II Controlled Substance (US) / Class A (UK)
| Active Ingredient | Oxycodone Hydrochloride 10 mg + Acetaminophen (APAP) 325 mg |
|---|---|
| Originator / Brand | Endocet (Endo Pharmaceuticals) / Percocet equivalent |
| Dosage Form | Yellow oval tablet imprinted E712 on one side and 10/325 on reverse |
| Controlled Schedule | DEA Schedule II Controlled Substance (US) / Class A (UK) |
|---|---|
| Primary Indication | Short-term management of acute moderate-to-severe pain requiring an opioid |
| Standard Dosing | One tablet every 4 to 6 hours as needed; maximum 6 tablets per 24 hours |
1. Mechanism of Action & Therapeutic Use
Synergistic dual-analgesic mechanism: oxycodone acts centrally on mu-opioid pathways while acetaminophen inhibits prostaglandin synthesis centrally and peripheral pain impulse generation.
2. Prescribed Administration Guidelines
- Take only as prescribed for acute short-term pain episodes.
- Do not exceed 4,000 mg total daily acetaminophen from all sources (prescription & OTC).
- Avoid combining with cold, flu, or headache remedies containing paracetamol.
- Take with food or milk if gastrointestinal nausea occurs.
Acetaminophen has been associated with acute liver failure, sometimes resulting in liver transplant or death, frequently linked to ingestion exceeding daily limits. Concomitant opioid risks include fatal respiratory depression and addiction.
3. Safety Precautions & Contraindications
Strict Contraindications: Hypersensitivity to oxycodone or acetaminophen; severe hepatic impairment; severe respiratory depression; acute asthma; bowel obstruction.
4. Adverse Reactions Profile
Common & Transient Effects:
Nausea, constipation, dizziness, somnolence, lightheadedness, diaphoresis.
Serious Reactions (Urgent Medical Evaluation):
Opioid overdose (respiratory depression, cyanosis), liver injury (upper-right abdominal pain, jaundice, dark urine).
5. Clinically Significant Drug Interactions
Alcohol markedly accelerates hepatotoxicity and respiratory depression. Benzodiazepines and sedatives compound lethargy.
6. Proper Storage & Security
Store at 20°C to 25°C away from moisture. Keep locked away from children and unauthorized individuals.
